Fig. 8.   ETA and ETB receptors promote mitogenesis in human mesangial cells. Quiescent cells were stimulated for 48 h with readdition of agonists (ET-1 and S6c at 100 nM) and antagonists (BQ-610 and PD-145065 at 100 nM) at 24 h. Cells were preincubated with ET receptor antagonists for 2 min before addition of ET-1 or S6c. Bromodeoxyuridine (BrdU) was present throughout the incubation, and BrdU incorporation into DNA was used to assess mitogenesis as described in MATERIALS AND METHODS and Ref. 40. Each bar represents >400 cells analyzed from 2 independent experiments. ** P < 0.01 vs. ET-1 alone by chi 2 analysis.
