Cover Cover: Images of superficial kidney cortex cysts from adult rats with autosomal-dominant polycystic kidney disease, acquired in vivo with the two-photon microscope. The cyst pictured in the background is derived from a proximal tubule, as evidenced by the accumulation of the secreted organic anion sulfonefluorescein (green fluorescence) in its cells. Accumulation of sulfonefluorescein, however, is uneven, with some cyst cells showing greatly diminished uptake, suggesting loss of the ability to transport this molecule. The cyst pictured in the foreground is probably derived from a distal tubule, as shown by the absence of cellular accumulation of sulfonefluorescein and the presence of bright blue nuclei (Hoechst 33342 dye) and little intrinsic fluorescence. See related article by Tanner GA, Sandoval RM, and Dunn KW. Two-photon in vivo microscopy of sulfonefluorescein secretion in normal and cystic rat kidneys. Am J Physiol Renal Physiol 286: F152-F160, 2004.
